PT-141: What It Is, How It Works & What the Research Shows
Overview
PT-141 (bremelanotide) is a melanocortin receptor agonist (MC3R and MC4R) that acts in the brain to increase sexual desire and arousal. Because it works through the nervous system instead of blood flow, it differs from drugs like sildenafil. It is FDA-approved as Vyleesi (2019) for hypoactive sexual desire disorder in premenopausal women, and earlier studies also showed effects in men. Nausea and temporary rises in blood pressure are the most common side effects.
Why researchers study it
Sexual desire
FDA-approved as Vyleesi for low sexual desire in women.
Arousal in men
Earlier studies showed effects on erectile response.
Brain-based action
Works through the nervous system, not blood flow.
How it works
Bremelanotide is a melanocortin receptor agonist. The precise mechanism responsible for improvement in HSDD is not fully established in its prescribing information.
Adamantane & the blood–brain barrier
Chemically attaching an adamantane group to a peptide can increase its lipid (fat) solubility. Researchers hypothesize that this modification may help certain compounds cross the tightly regulated blood–brain barrier. The effect depends on the specific modified molecule; greater lipid solubility alone does not establish brain delivery or clinical benefit. This does not apply automatically to the unmodified peptide and is not a reconstitution additive.
What the research shows
- Prescription bremelanotide has evidence for sexual desire and related distress in eligible premenopausal women with acquired, generalized HSDD.
Typical protocol
Reconstitute a 10 mg vial with 3 mL bacteriostatic water for 3.3333 mg/mL. Typical range: 0.5–2 mg as needed · as needed.
Common stacks
- PT-141 + Kisspeptin — Pairs central sexual-desire signaling with reproductive hormone support.
- PT-141 + Oxytocin — Pairs arousal signaling with bonding and connection signaling.
Potential benefits & side effects
Potential benefits
- Deeper, longer-lasting tan with less UV exposure (Melanotan)
- Increased sexual desire and arousal (PT-141, Melanotan II)
- Appetite suppression (Melanotan II)
Side effects
- Nausea (most common)
- Facial flushing
- Darkening of freckles and moles
- Temporary rise in blood pressure (PT-141)
- Spontaneous erections (Melanotan II, PT-141)
Lifestyle factors
- Start low: nausea is dose-related.
- Dose in the evening to sleep through nausea and flushing.
- UV: tanning still requires sun exposure — use sensible exposure and protection.
- Check moles regularly and see a dermatologist for any changes.
