PE-22-28: What It Is, How It Works & What the Research Shows

Overview

PE-22-28 is a synthetic analog of spadin, a peptide derived from the sortilin protein. It blocks TREK-1 potassium channels in the brain, a target linked to depression. In mouse studies, it produced antidepressant-like effects within days rather than weeks and increased the growth of new neurons in the hippocampus. It has not been studied in humans.

Why researchers study it

Mood

Produced fast antidepressant-like effects in mice.

Neurogenesis

Increased new neuron growth in the hippocampus.

TREK-1 target

Blocks a potassium channel linked to depression.

How it works

A spadin-derived peptide investigated for inhibition of TREK-1 and antidepressant-like effects in preclinical work.

Adamantane & the blood–brain barrier

Chemically attaching an adamantane group to a peptide can increase its lipid (fat) solubility. Researchers hypothesize that this modification may help certain compounds cross the tightly regulated blood–brain barrier. The effect depends on the specific modified molecule; greater lipid solubility alone does not establish brain delivery or clinical benefit. This does not apply automatically to the unmodified peptide and is not a reconstitution additive.

What the research shows

  • TREK-1 potassium-channel activity and antidepressant-like effects have been explored preclinically.

Typical protocol

Reconstitute a 10 mg vial with 3 mL bacteriostatic water for 3.3333 mg/mL. Typical range: 50–200 mcg 1x daily · daily.

Common stacks

  • PE-22-28 + Semax — Pairs a TREK-1 blocker with a BDNF-raising peptide for mood and cognition.

Potential benefits & side effects

Potential benefits

  • Improved focus and mental clarity
  • Reduced anxiety and stress reactivity
  • Better mood and motivation
  • Neuroprotective support (BDNF-related peptides)

Side effects

  • Nasal irritation with intranasal use
  • Mild headache
  • Overstimulation or irritability (Semax-type)
  • Drowsiness (Selank, DSIP)

Lifestyle factors

  • Cycle it: run on/off cycles to maintain responsiveness.
  • Timing: dose stimulating peptides (Semax family) early in the day; calming ones (Selank, DSIP) later.
  • Sleep and hydration have a bigger effect on cognition than any peptide.
  • Limit caffeine stacking to avoid overstimulation.
