Semax: What It Is, How It Works & What the Research Shows

Overview

Semax is a synthetic heptapeptide based on a fragment of ACTH (4-10), with a Pro-Gly-Pro tail added for stability, and it has no hormonal activity. It increases brain-derived neurotrophic factor (BDNF) and its receptor TrkB, supporting neuron survival and plasticity. Semax is approved in Russia for stroke recovery, cognitive disorders and optic nerve conditions, and is usually given intranasally.

Why researchers study it

Cognition

Explored for memory, learning and mental performance.

Neuroprotection

Used in Russia for stroke recovery; raises BDNF.

Focus & attention

Studied for attention and mental fatigue.

How it works

Synthetic peptide studied in neurological research; regulatory status varies by country.

Adamantane & the blood–brain barrier

Chemically attaching an adamantane group to a peptide can increase its lipid (fat) solubility. Researchers hypothesize that this modification may help certain compounds cross the tightly regulated blood–brain barrier. The effect depends on the specific modified molecule; greater lipid solubility alone does not establish brain delivery or clinical benefit. This does not apply automatically to the unmodified peptide and is not a reconstitution additive.

What the research shows

  • Neurological outcomes, neurotrophic signaling, and ischemic-injury mechanisms have been investigated.

Typical protocol

Reconstitute a 10 mg vial with 3 mL bacteriostatic water for 3.3333 mg/mL. Typical range: 200–800 mcg 1–2x daily (am/mid day) · 5 on 2 off or daily, 4–8 weeks.

Common stacks

  • Semax + Selank — The most common nootropic pairing: Semax for focus, Selank for calm.

Potential benefits & side effects

Potential benefits

  • Improved focus and mental clarity
  • Reduced anxiety and stress reactivity
  • Better mood and motivation
  • Neuroprotective support (BDNF-related peptides)

Side effects

  • Nasal irritation with intranasal use
  • Mild headache
  • Overstimulation or irritability (Semax-type)
  • Drowsiness (Selank, DSIP)

Lifestyle factors

  • Cycle it: run on/off cycles to maintain responsiveness.
  • Timing: dose stimulating peptides (Semax family) early in the day; calming ones (Selank, DSIP) later.
  • Sleep and hydration have a bigger effect on cognition than any peptide.
  • Limit caffeine stacking to avoid overstimulation.
