NAD+: What It Is, How It Works & What the Research Shows

Overview

NAD+ (nicotinamide adenine dinucleotide) is a coenzyme found in every cell. It is essential for converting food into energy and is used by sirtuins and PARP enzymes for DNA repair and cellular stress responses. NAD+ levels decline with age, which has driven interest in raising them. Most human research has studied precursors such as NR and NMN, which reliably raise blood NAD+; direct NAD+ infusions and injections are widely used in clinics but have fewer controlled trials.

Why researchers study it

Cellular energy

Essential coenzyme for turning food into ATP.

DNA repair

Fuels sirtuins and PARPs that maintain and repair DNA.

Healthy aging

NAD+ declines with age, driving interest in restoring it.

How it works

NAD+ acts as an electron-transfer cofactor and a substrate for enzymes such as sirtuins and PARPs. Intracellular biology and administered NAD+ are separate evidence questions.

Adamantane & the blood–brain barrier

Chemically attaching an adamantane group to a peptide can increase its lipid (fat) solubility. Researchers hypothesize that this modification may help certain compounds cross the tightly regulated blood–brain barrier. The effect depends on the specific modified molecule; greater lipid solubility alone does not establish brain delivery or clinical benefit. This does not apply automatically to the unmodified peptide and is not a reconstitution additive.

What the research shows

  • NAD is involved in cellular energy production and redox reactions.
  • NAD metabolism and supplementation are research areas.

Typical protocol

Reconstitute a 500 mg vial with 3 mL bacteriostatic water for 166.6667 mg/mL. Typical range: 25–200 mg 1x daily am · 2–4x week or daily, As long as needed.

Common stacks

  • NAD+ + MOTS-C — Mitochondrial support: NAD+ supplies the coenzyme, MOTS-C improves metabolic signaling.
  • NAD+ + SS-31 — Pairs NAD+ with a peptide that stabilizes the mitochondrial membrane.
  • NAD+ + Glutathione — Pairs energy-metabolism support with antioxidant support.

Potential benefits & side effects

Potential benefits

  • Improved energy and exercise capacity
  • Better insulin sensitivity and metabolic markers
  • Support for mitochondrial function
  • Potential fat-loss support (5-Amino-1MQ, AOD-9604, SLU-PP-332)

Side effects

  • Injection-site irritation
  • NAD+: flushing, chest tightness or nausea if pushed too fast
  • Fatigue or restlessness early in a cycle
  • GI discomfort with oral compounds

Lifestyle factors

  • Exercise: these compounds work alongside, not instead of, regular training — especially zone 2 cardio.
  • Sleep and circadian rhythm: keep a consistent sleep schedule.
  • Nutrition: whole-food diet with adequate protein; avoid chronic overeating.
  • Track bloodwork (glucose, lipids, liver markers) before and during longer cycles.
