5-amino-1mq: What It Is, How It Works & What the Research Shows

Overview

5-Amino-1MQ is a small-molecule (non-peptide) inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that is overactive in fat tissue. Blocking NNMT raises cellular NAD+ and SAM levels and shifts fat cells toward burning rather than storing energy. In diet-induced obese mice, it reduced body weight and fat mass and shrank fat cells without changing food intake. It has also been studied for preserving muscle stem cell function in aging mice. Human trials have not been published.

Why researchers study it

Fat metabolism

Blocks NNMT, shifting fat cells toward burning energy in mice.

NAD+ levels

Raises cellular NAD+ by freeing up its building blocks.

Muscle aging

Studied for preserving muscle stem cell function in aged mice.

How it works

A small-molecule NNMT inhibitor studied for changes in nicotinamide metabolism and cellular energy regulation, primarily in preclinical models.

Adamantane & the blood–brain barrier

Chemically attaching an adamantane group to a peptide can increase its lipid (fat) solubility. Researchers hypothesize that this modification may help certain compounds cross the tightly regulated blood–brain barrier. The effect depends on the specific modified molecule; greater lipid solubility alone does not establish brain delivery or clinical benefit. This does not apply automatically to the unmodified peptide and is not a reconstitution additive.

What the research shows

  • NNMT inhibition and cellular energy metabolism are research targets.
  • Adiposity and metabolic changes have been investigated in preclinical models.

Typical protocol

Reconstitute a 50 mg vial with 3 mL bacteriostatic water for 16.6667 mg/mL. Typical range: 2.5–5 mg once daily.

Common stacks

  • 5-Amino-1MQ + NAD+ — Both aim to raise cellular NAD+ through different routes: NNMT inhibition and direct supply.
  • 5-Amino-1MQ + MOTS-C — Pairs NNMT inhibition with AMPK activation for metabolic support.

Potential benefits & side effects

Potential benefits

  • Improved energy and exercise capacity
  • Better insulin sensitivity and metabolic markers
  • Support for mitochondrial function
  • Potential fat-loss support (5-Amino-1MQ, AOD-9604, SLU-PP-332)

Side effects

  • Injection-site irritation
  • NAD+: flushing, chest tightness or nausea if pushed too fast
  • Fatigue or restlessness early in a cycle
  • GI discomfort with oral compounds

Lifestyle factors

  • Exercise: these compounds work alongside, not instead of, regular training — especially zone 2 cardio.
  • Sleep and circadian rhythm: keep a consistent sleep schedule.
  • Nutrition: whole-food diet with adequate protein; avoid chronic overeating.
  • Track bloodwork (glucose, lipids, liver markers) before and during longer cycles.
